How does Avil Syrup work?

• Pheniramine, the active ingredient in Avil, is a histamine H1-receptor antagonist. It also has anticholinergic and moderate sedative effects.

• Common side effects include sedation, dizziness, tinnitus, insomnia, nausea, vomiting, dry mouth, constipation, and urinary retention. Rarely, it may cause blood dyscrasias like agranulocytosis or hemolytic anemia.

Indication of Avil Syrup

Avil Syrup is an antihistamine medicinal drug that contains Pheniramine as the main element present in it. This medication acts as a histamine antagonist that is used for the treatment and prevention from diseases such as hay fever, allergic rhinitis, pruritus, and allergic dermatoses. Pheniramine is also used to control common cold, flu, allergies, watery eyes, running nose, itchy throat, and breathing troubles.

Pharmacology

Pheniramine works by competing with histamine for the H1 receptor and acts as an inverse agonist once it binds. This action leads to a decrease in H1 receptor activity, which in turn reduces itching, vasodilation, and capillary leakage, ultimately leading to less redness and swelling. This effect is particularly evident in the suppression of the histamine-induced wheal (swelling) and flare (vasodilation) response.

In the central nervous system (CNS), the inverse agonism of the H1 receptor by pheniramine is responsible for the sedative effects commonly associated with first-generation antihistamines. Additionally, pheniramine's binding to H4 receptors and its inverse agonism at these sites may also help reduce itching by counteracting inflammation.

Dosage & Administration of Avil Syrup

Adult Dose: For allergic conditions, the recommended dose is 15-30 mg of syrup, 2-3 times daily. The maximum dose should not exceed 3 mg/kg/day. For motion sickness, the same dosage applies, and the first dose should be taken at least 30 minutes before traveling.

Child Dose: For children aged 5-10 years, half a 45 mg tablet up to 3 times daily is recommended. For children over 10 years, the dosage is 15-30 mg of syrup, 2-3 times daily. The maximum dose should not exceed 3 mg/kg/day. The first dose for motion sickness should be taken at least 30 minutes before traveling.

Interaction

Avil may mask the ototoxicity caused by aminoglycoside antibiotics. It may also potentiate the CNS depressant effects of alcohol, sedatives, opioids, and other similar drugs. MAOIs, atropine, and tricyclic antidepressants (TCAs) may enhance its antimuscarinic effects.

Contraindications

Avil should not be used in patients with symptomatic prostatic hypertrophy, newborns, or premature infants.

Pregnancy & Lactation

Avil may impair your ability to drive or operate machinery. It has the potential for abuse and should be used with caution in patients with narrow-angle glaucoma, asthma, or severe cardiovascular disease. Its antiemetic effect may mask the symptoms of other conditions. Caution is advised in pregnant or breastfeeding women, as well as in the elderly.

Precautions & Warnings

• Avoid using Pheniramine Maleate if you have prostatic hypertrophy (enlarged prostate gland).
• Do not use Pheniramine Maleate if you are currently taking MAO inhibitors.
• Inform your doctor if you have conditions such as narrow-angle glaucoma, asthma, severe cardiovascular disease, or kidney, or liver problems.
• Consult your doctor before using Pheniramine Maleate if you are pregnant or breastfeeding.
• This medication may cause drowsiness. Only drive or operate machinery if you are fully alert.
• Pheniramine Maleate is not recommended for children under 5 years of age, newborns, or premature infants.
• The anti-emetic effect of Pheniramine Maleate may mask symptoms of other conditions. Always keep your doctor informed about your full medical history.
